![]() Rather than it being about Susan’s life falling apart, its focus is that of her rediscovering her will to live. The Cat Lady is dark, to be sure, but the game has a noticeable element of hope to it. Downfall is very different from The Cat Lady, and not just in terms of graphics and interface, but in tone as well. Now I suppose I could’ve made this a footnote when discussing The Cat Lady, except I’d be doing all of you a disservice by glossing over it like that. ![]() The Cat Lady is sort of the “spiritual successor” of Downfall. The Cat Lady and Downfall were produced by the same man, Remigiusz Michalski. The game is available on Desura, GOG, and Steam for just $9.99. Top it all off by the fact that The Cat Lady has an awesome soundtrack from micAmic, and I’m just not sure how you could pass this title up. The game features full voice-acting, and if you’re familiar with online animation at all, you’ll be delighted (or unnerved, take your pick) to learn that David Firth offered his unique voice-acting talents to this game. This indie title relies mostly on inventory puzzles, but the story is quite entertaining and the characters rather likeable. The catch? In order to keep her new life, Susan has to kill five evil “parasites” - aka, psychopaths. In the afterlife, she meets a strange old woman who resonates with menacing power, and this person offers Susan a second chance at life. You play as Susan Ashworth, an extremely depressed woman who, at the start of the game, attempts suicide. The story has tremendous heart and it gets a plus one for managing to present a protagonist that we wouldn’t expect. It felt like some gothic horror mind trip from the 90s. I’ve played it several times since because I loved the catharsis that came with its story. I remember when this game first came out back in December 2012 and all the buzz it generated. Giger’s work, I might add) but if you’re a masochist I suppose you can give that a try as well. The sequel to this game may have been a much less stellar FMV mess ( without the benefit of H.R. The Macintosh version is also fairly decent. My suggestion is to use AmigaForever to emulate it, as the Amiga version is probably the best in terms of graphics and sound quality. The game is now abandonware, so you can download it for free. Creepy and crazy? You betcha! Giger’s work was stellar for this game, achieving a level of design many titles were envious of at the time. ![]() On the first night he stays in the house, he has a nightmare that some alien machine injects an embryo into his brain. You play the game as Mike Dawson, an ad executive and writer who has just bought an old mansion. Giger himself did work for? Dark Seed features ornate, but menacing art. So why not start this list off with a game that H.R. Giger had on Tormentum: Dark Sorrow’s artwork. Here on Cliqist, we easily saw the influences H.R. I would be remiss if I didn’t mention this horror adventure classic. That’s why I’ve compiled a list of nightmarish games that are similar to it! Here they are, in no particular order of preference: As a fan of metal music and horror films, I doubly appreciate such game titles. I always felt that adventure games were ideal for things like that given the nature of interaction and the strong focus on story. The elements of horror in Tormentum crept under your skin, making you feel unease. It explores the idea of redemption, but not just that, how we perceive morality at all. Tormentum: Dark Sorrow ( review / strategy guide) is unique as a casual-level adventure game because of its beautiful, dark, and surreal imagery.

![]() There are 23 pairs of chromosomes in a diploid human body cell. Only two pairs of chromosomes are shown in the diagrams below. The stages of mitosis are: prophase, metaphase, anaphase and telophase. The cell membrane pinches in and eventually divides into two daughter cells. New membranes form around the chromosomes at each end of the cell. The membrane around the nucleus disappears.Ĭhromosomes and their copies line up in the middle of the cell.Ĭhromosomes and their copies are pulled to different ends of the cell. The DNA in chromosomes and their copies condenses to become more visible. The DNA in chromosomes copies itself ready for mitosis. The cell spends most of its life in this phase. Every base pair of their DNA is identical. The daughter cells are clones of each other. Microtubules from one centrosome attach to the kinetochore (protein structures at the centromeres) of one chromosome from each of the homologous pairs, while the other centrosome connects to the kinetochore of the other chromosome in each homologous pair, and each homologous pair moves towards the metaphase plate (where they line up before anaphase).Mitosis is a type of cell division in which a diploid body cell copies itself and finally divides into two identical diploid daughter cells. Centrosomes move to opposite ends of the cell, and the nuclear envelope dissolves.ħ. This formation occurs because of sister chromatid cohesion, where a gene that has been given to the homologous pair in synapsis is still bonded to the corresponding part on the sister chromatid of its former chromatid.Ħ. Some of these homologs have one or more chiasmata, an X shaped region where a genetic rearrangement has occurred. Synapsis ends, and the homologs move slightly apart, no longer bonded along their lengths like in the synaptonemal complex.ĥ. This forms a synaptonemal complex.ģ.The random rearrangement of corresponding genes occurs between the non sister chromatids (because at this stage each chromosome consists of two sister chromatids).Ĥ. Ans: The \(6\) stages of the cell cycle are interphase, prophase, metaphase, anaphase, telophase, and cytokinesis. The paired homologs become physically connected along their lengths through a process called synapsis. Chromosomes condense and homologs loosely pair along their lengths, aligned by gene.Ģ. The above is also how a 46 chromosome (double strand) cell in mitosis can result in 2 daughter cells each with 46 chromosomes (single strand).Įven Sal admits how confusing this is, but he explains all this visually in a separate video differentiating the terms chromatid, chromosome, and chromatin.ġ. So a haploid parent cell of 23 chromosomes (double strand) just created two haploid daughter cells of 23 chromosomes (now single strand). When the chromatids are separated they are now called chromosomes Anaphase II splits the sister chromatids which now separate (23 chromatids go to one pole and 23 chromatids go to other pole). The parent cell starts with 23 chromosomes (EACH double stranded=two sister chromatids, so there are 46 chromatids. This means that in meoisis II when we split the sister chromatids: It is only when the sister chromatids separate are they each considered separate chromosomes. The single strand chromosome (1 chromosome) became two stranded yet attached identical sister chromatids (still 1 chromosome) Likewise chromosome 5 of dad is similar to chromosome 5 of mom)Īfter replication how many chromosomes do we have? In interphase before S (replication phase) we have 46 single stranded chromosomes: 23 are from mom and 23 are from dad (they code for the same things meaning chromosome 1 of mom codes for the same thing as chromosome 1 of dad. ![]() Remember that when replicating in interphase, the chromosome number DOES NOT CHANGE ![]() No matter the style, all Daggerwing eyewear utilize Carl Zeiss optical lenses to provide the best and clearest eyewear for your vision, including prescription shades and glasses.Įxpress your inner retro look in the modern era with our female shades. ![]() These unique female designer shades offer a fun luxury look from styles from the 1950’s leading up to the modern era of polarized and tinted styles. AM cells preferentially accumulated in blood of patients with severe ABMR manifestations including late-onset of rejection post-transplant, presence of IgG3 DSAs, and increased vascular inflammation in kidney allografts. IL-21 was a potent inducer of AM cells, their upregulation of T-bet and IL-21R, and promoted their differentiation into DSA-producing plasma cells. AM cells in patients undergoing ABMR markedly upregulated IL-21R, and the downstream transcription factors, IRF4 and Blimp1, displayed a plasma cell precursor transcriptional profile and expressed more restricted IGHV sequences, reflective of clonal expansion, when compared to other memory B-cell subsets ( p < 0.001). AM cells were less frequent in DSA+ABMR patients and at baseline levels in DSA patients ( p < 0.001). Results: We identified expanded numbers of CD27 +CD21 - activated memory (AM) B cells that expressed the transcription factor T-bet in blood of patients who developed DSAs and progressed to ABMR. There were 48 patients without DSAs of those with DSAs, ABMR emerged in 20 patients, but not in 28 patients. ![]() Methods: In a cohort of 96 kidney transplant recipients, we performed 22-color spectral flow cytometry, RNA-seq and in vitro assays to profile circulating B cells, as well as multiplex immunofluorescence and RNA-seq to profile infiltrating B cells in allograft biopsies. ![]() Yet, the cellular states underlying alloreactive B-cell responses and the molecular components controlling them remain unclear. ![]() Starzl Transplantation Institute, University of Pittsburgh Medical Center, Pittsburgh, Pennsylvania, USAīackground: Donor-specific antibodies (DSAs) and ensuing development of antibody-mediated rejection (ABMR) are detrimental to organ transplants. This process deadens the language, and the result in the case of the Harper’s letter is a graveyard of prose, without a single pungent phrase or sentence worthy of quotation. They can bear no traces of their individual authors, and the easiest way to scrub those traces is to write in a numbing, anonymized style, free of idiosyncrasy and wit. Open letters tend to be composed inclusively, so as many people as possible will sign them. Here are reasons why the genre of open letters should die: ![]() Open letters are terrible, and you should never write one or sign one. I told the letter’s organizers that if I have something to say, I will write my own damn letter. But I never sign petitions or open letters. Perhaps because I spend a lot of time listening to people with crazy opinions, I am sympathetic to the view that the only way to live a healthy intellectual life is to expose oneself constantly to weird or detestable opinions. The signers argued that you should keep that category of the undebatable as small as possible, and not add to it whenever you form an opinion. We all have limits some topics are beneath our dignity to debate. The letter warned of “ideological conformity” and “illiberalism” in liberal institutions, and it noted a tendency to confront dissenting opinions not with debate but by going after the job of the dissident, or even by going after the job of those who merely note the existence of the dissent. Last month, I was asked to sign the “Letter on Justice and Open Debate,” now published in Harper’s with the signatures of 153 journalists and academics, including contributors to The Atlantic. ![]() What dog doesn’t like a treat! Dogs like to sniff things out and although their sense of smell may also deteriorate as they age, try hiding treats around the house and garden for them to sniff out. It’s worth bearing in mind that older dogs may have sore spots that they want to protect, or get tired quicker than younger playmates, and can show signs that they want some space such as growling at other dogs. Keeping them on a leash will also help you control their interaction with other younger dogs who want to play chase. You may find that if their eyesight or hearing is impaired, you need to keep them on a long leash so that they don’t wander off and cannot hear you recall them. Physical and mental stimulation are just as important for more senior poochesĬontinue to walk your dog regularly, but consider taking them for more frequent, shorter walks instead of a long, strenuous walk.If you have played with your pooch since they were a puppy, it will continue to be something they enjoy as they get older, but you may need to adapt playtime games to make them more gentle but just as stimulating. But we also need to be aware that a dog’s behavior changes as they get older, and although you should continue to exercise and play with them, you will need to adjust the form of play accordingly in order to keep your best friend mentally and physically stimulated. We are used to playing with puppies and helping them to understand their new environment through training and constructive play. You may even see signs of muscle weakness or joint pain. Energy levels might reduce, theirs eyesight might deteriorate and they may suffer some hearing loss. However, like humans, all dogs are individuals and some dogs can still be very lively as they grow older, whereas others may be prone to signs of old age earlier. If dogs don’t get regular exercise, they could become anxious or frustrated, their physical and mental condition could deteriorate and they may get destructive at home out of boredom.Īlways keep your dog’s health front of mind as the years pass. It’s a great way to release any tension they may have and will enforce the bond between you. Not only is it good exercise, play provides mental, physical and social benefits for dogs. Seeing your pooch chasing a ball or frisbee, playing tug-of-war with a rope or chasing and playing with other dogs is a sight for sore eyes - but the importance of continuing playtime with your dog when they get older cannot be underestimated. Remember that you do not need to duplicate the reference photo you are using.Ĭreating art is all about how you see the subject and paint it with your style. It is also essential to work on your color mixing and understand how to create natural-looking colors on your painting surface. Especially if you are using acrylics as your painting medium, consider the following factors for you to succeed in this project. It takes patience and a keen heart to learn a new skill. But rest assured that once you have tried working with your waves, you will discover that they are fun, allowing you to enhance your skills better than before. Special thanks to Diane Griffiths for sharing these tips for painting water! To see more of Diane’s seascapes and coastal paintings, please visit waves with acrylics may seem challenging for artists who have never been to this type of design. Use a ruler if it helps! I also like to turn my painting upside down to spot those issues. Lastly, check your reflections to ensure they’re directly below the reflected object. It’s all too easy to overdo, however, so pick your knife strokes carefully. Water moves constantly, and this will help to create the impression of movement with just a few strokes of the knife. The crisper edges of colour created by a palette knife make for a great water reflection. Try using a palette knife yourself to blend and merge colours together when painting water. ![]() For reflections I’ll often swap to juicy thick oils and a palette knife to get the effect created in At the Foot of the Fern Pit. In a painting, reflections in the water can be like a mirror image or it could be just a suggestion of the world above it reflects. (We think of water as blue, but it’s only blue because it’s often reflecting a blue sky). But don’t let that shape your decisions-sometimes I really do want my reflections to be equally colourful it just depends on my composition.Īnd don’t be afraid to use any colour in your reflections-even if it isn’t one traditionally found when looking at water. the darks will be slightly lighter and the lights will be slightly darker. I’ve always read that reflections are more muted than the original i.e. When adding reflections in the water, take some liberties. If I’m doing water highlights on the sea, I’ll use a brush to gently splash some speckles over the water and that works brilliantly to pull out little highlights of the sun. Instead, find ways to let your tools do the work for you. I don’t think it’s worthwhile to try to capture every wave and ripple-it will drive you insane. I then add just a bit of white to give a sense of waves crashing onto the beach. It’s a bit of a gamble doing it this way, but you can get some great, watery effects. I won’t let it drip exactly, but I will allow it to find its way across the canvas. ![]() Sometimes I’ll use heavily pigmented acrylic paints mixed with water so it can be use like watercolour and I let the water take over. I do a lot of experimentation as I paint, but I also have some techniques that I keep going back to: 1. ![]() You can see this in my paintings Watching Boardmasters, Fistral Beach shown at the beginning of this article, and Crantock Beach below.Įach time I look at them, I’m transported to summer holidays-that’s the type of “special” feeling I’m going for. It also captures a feeling or memory of being on that beach. It’s hard to explain exactly why a photo might feel that way, but you know it when you see it, and it makes painting the scene much more enjoyable and interesting.įor example, recently I’ve started a new collection of beach paintings which verges on the abstract, but it captures the reality of what you see on a busy summer’s day on the beach. I just like to make full use of my artistic license!Īs a general tip, if you paint from photos, try to choose ones that are truly special-my favorite photos are the ones where something extraordinary jumps out at me, or where there’s something ordinary-but-meaningful, and it tugs on the heart strings. I might try to bring out more intense colours, exaggerate the waves, or change the colours to fit my current colour palette. I use what I see in front of me in order to understand what I’m painting and how the light reflects and refracts through water, but. This allows me to have the best of both worlds when painting. but I do create a lot of paintings around the coast, so I need to be able to paint the wet stuff!Īs I paint, I tend to follow the principle that nothing is completely observed and nothing is completely imagined. I don’t consider myself the strongest painter of water.
